A toxicidade do manganês em humanos é um risco ocupacional bem conhecido para pessoas que inalam o pó de manganês. Altas concentrações de manganês circulante como resultado da nutrição parenteral total também foram associadas à toxicidade do manganês. Pessoas com doença hepática crônica têm patologia neurológica e sinais comportamentais de neurotoxicidade do manganês, provavelmente porque a eliminação do manganês na bile está prejudicada. O efeito mais proeminente é a patologia do sistema nervoso central, especialmente no sistema motor extrapiramidal. As lesões e sintomas são semelhantes aos da doença de Parkinson.

Manganese toxicity in humans is a well-recognised occupational hazard for people who inhale manganese dust. High concentrations of circulating manganese as a result of total parenteral nutrition have also been associated with manganese toxicity. People with chronic liver disease have neurological pathology and behavioural signs of manganese neurotoxicity, probably because elimination of manganese in bile is impaired. The most prominent effect is central nervous system pathology, especially in the extra-pyramidal motor system. The lesions and symptoms are similar to those of Parkinson’s disease.
